Bovine herpesvirus type 1 in cumulus-oocyte complexes collected from naturally infected cows PAB

Abstract: The objective of this work was to investigate the presence of bovine herpesvirus type 1 (BoHV-1) in follicular fluid and in cumulus-oocyte complexes (COC) recovered from naturally infected cows but with no clinical signs of the disease. Cows that were seropositive (n=38) or seronegative (n=8, control) to infectious bovine rhinotracheitis were selected after a serum neutralization test in microplates. The presence of the virus was investigated by PCR in COC and in follicular fluid. Viral DNA was not found in any of the samples. The obtained results suggest that serologically positive cows with no clinical signs of the disease offer negligible risk of transmitting BoHV-1 by COC or follicular fluid.

Classification of Bovine Follicles Based on the Concentrations of Steroids, Glucose and Lactate in Follicular Fluid and the Status of Accompanying Follicles OAK

A simple and clear means to identify the physiological status of follicles is essential for study of follicular biology. In the present study, we verified a novel classification procedure based on analysis of the follicular population and glucose concentration in follicular fluid (FF) as an alternative method to classify bovine follicles. Paired ovaries were collected from heifers, and the number of follicles and stage of the CL were recorded. Follicles were initially divided into the following 3 groups according to diameter and the ratio of E2 and P4 (E/P): E2 active (E-A: E/P≥1), E2 inactive (E-I: E/P<1, ≥8.5 mm) and small follicles (E/P<1, <8.5 mm). Effect of charcoal-treated bovine follicular fluid on the secretion of FSH in ovariectomized and intact prepubertal heifers Braz. J. Vet. Res. Anim. Sci.

The aim of the experiment was to determine whether charcoal-treated bovine follicular fluid (bFF) removed from visible (< 22 mm) follicles altered the secretion of follicle-stimulating hormone (FSH) in intact and ovariectomized prepubertal heifers. After a 10-ml injection of bFF given at three consecutive 8-h intervals, secretion of FSH was depressed in approximately 44% of the ovariectomized heifers but there was no effect in intact heifers. When the bFF treatment ceased, there were no rebound effects on FSH concentrations above that of controls pretreatment levels. Ten multiparous non-lactating cows were randomly assigned to one of two groups. On days 8 to 12 after estrus, the first group was given 100 ml antiserum against steroid-free bovine follicular fluid (anti-bFF), which was produced in ovariectomized sheep. The second group (control) was given 100 ml nonimmune ovariectomized sheep serum. Six hours after injection, both groups were superovulated with FSH (18 NIH-FSH-S1 units) and LH (0.29 NIH-LH-S1 units) in a 4-day decreasing regime. To evaluate the effect of follicular fluid on in vitro maturation, pig oocytes were cultured in the presence of hormones where 10% of fetal calf serum (FCS), 10% of follicular fluid from large follicles (l-pFF), 10% of follicular fluid from medium follicles (m-pFF) or no supplement were added. When oocytes where matured in medium containing the hormones the addition of different supplements did not affect (P<0.05) nuclear maturation. However, changing the supplement altered the cytoplasmic maturation, with higher rate (P<0.05) observed in the l-pFF group. The activity of α3-D-mannoside-β-1,2-N-acetylglucosaminyltransferase I (GnT I; EC 2.4.1.101), which catalyzes the first step in the conversion of oligomannose to complex or hybrid N-glycans of glycoproteins, was detected in bovine follicular fluid (bFF). The GnT I activity in bFF had a pH optimum of 5.8 and an absolute requirement for either Co2+, Mn2+, or Mg2+, the activity being stimulated by these cations in the above order. The apparent Km value for α1-3α1-6 mannopentaose of GnT I in bFF was 2.17 mM. The substrate specificity for the GnT I activity decreased in the following order: α1-3α1-6 mannopentaose> α1-3α1-6 mannotriose>α1-3 mannobiose.
